The movie wouldn't necessarily have to take place in space. They could do another invasion attack, perhaps this time from a global scale to differentiate it from the first movie.
No reason why Warlock or the Silver Surfer would have to be included. Same for the FF. They can still tell a great story without those characters.
I don't think Thanos will be in the next Avengers movie as the primary villain. Thanos isn't a typical Avengers villain. He's a cosmic character whereas the Avengers are more Earth-bound. There are other options for the Avengers to fight from their own pool of villains, such as the Masters of Evil, the Red Skull, Kang, and Ultron. As a trend, the after credit scenes mostly set up another next Marvel movie and not the sequel. For the after credit scenes, Iron Man 2 set up Thor, Thor set up the Avengers and Captain America, and Captain America set up the Avengers. In this regard, it's possible that Thanos's post credit scene is going to set up another movie, such as Guardians of the Galaxy, something that Marvel has stated they want to do.
